PHYSICAL AND CHEMICAL MUTAGENESIS IN CATHARANTHUS ROSEUS PLANT USING SCOT MARKERS

چکیده

The current study comprising molecular identification of the mutagenic effects gamma rays and sodium azide on Catharanthus roseus using scot markers took place at Department Biology, University Kufa, Iraq. seeds two cultivars, i.e., ‘victory carmine’ a local variety (pink variety) L (carmine var.) received irradiation with doses (50 Gray 75 Gray). Afterward, these got soaked in four concentrations (0.2%, 0.4%, 0.6%, 0.8%) to induce genetic variation cultivar. non-treated served as control treatments for comparison treated ones. Accomplishing was an average 18 Gy/h Cobalt-60. treatments’ evaluation were through 10 start codon targeted (SCoT) DNA markers. higher number distinctive fingerprints three produced by primers SCoT-26, while primer SCoT-60 provided distinct fingerprint only one treatment. However, SCoT-40 failed recognize any treatment fingerprint. highest size (2488 bp) came from SCoT-54, whereas lower (143 resulted SCoT-12. Primer SCoT-33 value chief unique bands, reaching 33 18, respectively. amplified bands 77 SCoT-54. monomorphic SCoT-12 gave polymorphic polymorphism, discriminatory (15, 68.18, 20.83, respectively), SCoT-9 efficiency.

The seco-iridoid pathway from Catharanthus roseus

The (seco)iridoids and their derivatives, the monoterpenoid indole alkaloids (MIAs), form two large families of plant-derived bioactive compounds with a wide spectrum of high-value pharmacological and insect-repellent activities. Vinblastine and vincristine, MIAs used as anticancer drugs, are produced by Catharanthus roseus in extremely low levels, leading to high market prices and poor availab...

5-Methyltryptophan resistant cells of Catharanthus roseus.

Several cell lines resistant to 5-methyltryptophan were selected from wild type cells of different Catharanthus roseus suspension cultures. The resistant cells had up to 30 times the normal levels of free tryptophan. Despite the increased pool size of tryptophan anthranilate synthetase activity of resistant cells was as sensitive to inhibition by L-tryptophan as wild type cells. The overproduct...

Identification of molecular markers for flower characteristics in Catharanthus roseus producing anticancer compounds

Catharanthus roseus or vinca produces over 130 TIAs with pharmaceutical value. Two of them, namely vinblastine (VB) and vincristine (VC), are species-specific and extensively used in anticancer chemotherapeutics. To detect C. roseus genotypes abundant in desired TIAs, contrasting genotypes should be analyzed via marker-assisted selection (MAS) to avoid analytical quantitation. A trustable morph...
